Is it possible to generate a pulse output based on either the rpm or vehicle speed?

Yes, depending on the ECU.
If it's the S100 in your signature, then you can generate them on AUX1, 2 and 3. Aux 3 can generate either a fixed 50% duty cycle with altering frequency, or a fixed frequency with different duty cycles.

I can see Aux3 can generate a varying frequency output from a number of inputs.

I need to send out an engine and road speed signal to the power steering controller and a road speed to trip meter. They will all need a variable frequency output, but it appears that only Aux3 can do this. Aux1 and Aux2 produce a selectable but fixed frequency and variable amplitude output - am a reading the manual correctly?

jaycar offer this, which allows to receive a pulsed signal, and adjust it +/- 100% as needed.

It might be easier to take an existing signal and use it instead
